More fuel is always better than less.

is the 255 in tank suffice enough for the 42lbers?

Definatley want to go with at least 42's. The 255 should support about 450-500 rwhp "every car/combo is different" and if You want to be safe or possibly save a yourself a trip back to the dyno then get an inline pump or BAP.

will I need to get this combo tuned? if so how much will a decent tune run me?

SCT chip & dyno tune will be needed. Price will depend on the car's running condition which will reflect on the amount of dyno time You need to get the tune rite.

Tuning is usually done by the hour. I think a good tune will run between $400-$500 including the chip. Call AED they will take care of you
I would go with 42's, a 255lpr pump, and SCT MAF. You should have enough fuel.
The best thing to do is find a tuner and get their recommendation. This will make the tuning process a lot smoother
