I"m trying to start over with a computer I've had for a few years and I want to reformat the C: drive and reinstall Windows. It also has a slave drive E:. If I save all my files I want to keep to the E:, will all those files stay safe and not be lost when i reformat the C: drive? After I get this done, what anti virus and spyware do you recommend? Thanks

Make sure you reformat and install windows on the correct partion/ hard drive (C) and you will be fine.

You should back up all of your files in case you press the wrong button and reformat your E:

Really low odds of this happening, but I would say that you do it just in case.

you should be good, if you do that. You could also disconnect the drive too as to avoid any overwrite. As for A/V I use AVG the free version with no issues. In the office we have a enterprise solution but for home use AVG should be good and you can get it from www.filehippo.com. Also grad spybot and super anti-spyware and maybe AdAware. All of them are free from the link I posted.
